Hereinwe reportan optimizedsynthesisfor dolichyldiphosphochitobiose(GlcNAc2-PP-Dol25), a probeusefulforbiochemicalandstructuralstudiesof proteinN-glycosylationin eukaryoticcells.Weimprovedthreephosphatecouplingstepsin termsof yieldsandreactiontimes,namelychitobiosephosphorylation,dolicholphosphorylation,andphosphate-phosphatecoupling,by adjustingreagentsandconditions.We alsodevelopedan efficientpreparativereverse-phaseHPLCpurificationprotocolfollowedby ion exchangestepto obtainthepureproductas a stablesodiumsalt.Theseoptimizedproceduresensurea reliablesupplyof GlcNAc2-PP-Dol25for enzymaticstudies.
