Mass Spectrometric Investigation of the Interaction of Metallocenes with Biomolecules

Bent Metallocenes (Cp2MCl2) have fond wide interest as anticancer agents with DNA as possible target. One serious drawback is the low solubility in water and, in case of titanocene, the instability to hydrolysis of the cyclopentadienyl ligand at physiological pH leading to inactivation. In recent literature, cyclodextrins and cucurbiturils are proposed as host molecules to overcome these drawbacks, especially to protect the Cp ligands in titanocene from getting lost. These findings are based mainly on molecular modelling, solid-state structure investigation, and NMR measurements. Therefore, mass spectrometry may give complementary insight into the benefit of such host guest structures as drug delivery systems.
