Potential (trifluoromethyl)diazirine-basedTRPML1 ion channel ligands were designed and synthesized,and their structures were determined by single-crystal X-raydiffraction analysis. Photoactivation studies via19F NMR spec-troscopy and HPLC-MS analysis revealed distinct kineticalcharacteristics in selected solvents and favorable photochemicalproperties in an aqueous buffer. These photoactivatable TRPMLactivators represent useful and valuable tools for TRPMLphotoaffinity labeling combined with mass spectrometry
