Every day we experience episodes, which we encode effortlessly on the fly by forming what, where and when associations.
A recently published experiment of our research group on vocabulary learning during deep sleep suggests that hippocampus-mediated rapid relational encoding is even feasible in a state of unconsciousness such as deep sleep (Züst, Ruch, Wiest, & Henke, 2018).
Since hippocampal activity climaxes during up-states of cortical slow-oscillations and slow-oscillations result from alternations between phases of high neuronal activity and plasticity (up-states) and phases of neuronal silence (down-states), our study tested, if the slow-wave phase is crucial for de-novo learning during sleep.

We aimed at enlarging up-states to facilitate sleep-learning using a closed-loop procedure, in which we targeted acoustic stimuli to up-states (versus down-states: control condition) of slow-oscillations during deep sleep. Pairs of foreign words and German words were played for associative learning to the sleeping participants. Memory for sleep-played vocabulary was assessed following waking using an implicit memory test.

Together, this experiment aims to investigate whether sleep-learning (1) is mediated by slow-oscillation up-states, and (2) can be enhanced with acoustic stimulation that increases slow-oscillation amplitudes.
